Complexes of aminoclays and polyelectrolytes

This thesis deals with the preparation of aminoclay and its products resulting from the interaction of aminoclay with natural polyelectrolyte. The theoretical part deals with the study of aminoclay and selected polyeletrolytes, which were used in the experimental part of the thesis. Potassium pectate, sodium alginate and sodium hyaluronate were chosen as polyelectrolytes. The aim was to let these two groups of compounds interact and to observe the formation of an aminoclay polyelectrolyte complex, taking form of a gel or a precipitate when appropriate concentrations were chosen. The resulting products were characterized in appearance and pH of the products was measured.
Complexes of aminoclay and selected organic compounds

This paper describes the preparation of aminoclay and complexes of aminoclay with selected bioactive substances. The theoretical part deals with the study of aminoclay and substances suitable for complexes and suitable modification of preparation these complexes. Experimental part of this paper consists of preparation of aminoclay complexes and verification of their formation. In order to verify the formation of the complex by binding of bioactive compounds to the free amino groups of aminoclay were used methods FT-IR (Fourier transform infrared spectroscopy) and XRD (X-ray diffraction). For selected substances, binding to the aminoclay would improve their properties, which was the main motivation for the research. The resulting complexes could be more effective by replacing existing substances used in the pharmaceutical industry and biomedical applications.
Phase separation in the system of aminoclay-biopolymer

The aim of this diploma thesis was to study the structure and to evaluate the properties of the hydrogel prepared by the interaction of the aminolclay with the biopolymer. Representatives of the biopolymers were selected from low to medium molecular weight sodium hyaluronate and sodium polystyrenesulfonate. On the basis of the experiments carried out, it was found that phase separation takes place only when the aminolclay interacts with medium molecular weight hyaluronan (MMW). In the experimental part, analyzes of this sample were carried out in order to determine the formation of phase-separated hydrogel by influencing the solution by ionic strength, investigation of hydrogel extinction in various organic solvents, stability of hydrogel under extreme temperature conditions, the effect of storage on its degradation, etc. Viscoelastic properties were experimentally proven by rheology and thermal analysis detected binding water. Inhibition of microorganisms was confirmed by antibacterial diffusion assays. All experiments were carried out for the use of the Aj-HyA hydrogel (MMW) in the field of medical applications, specifically for the modern method of wet wound healing of the skin.
Interactions of aminoclays with natural polyelectrolytes

This thesis deals with the preparation of products of interactions of aminoclay with natural polyelectrolytes. Sodium hyaluronate was chosen as the natural polyelectrolyte. The concentration and volume ratio of solutions of aminoclay and natural polyelectrolyte which led to the formation of any type of product were studied. The resulting coagulates were characterized by microscopy, FTIR spectroscopy and rheology. Furthermore the possibility of interactions with fluorescent probes was studied.
Adsorption of biologically active substances in colloidal solutions of aminoclay

This paper builds on a previous study of magnesium aminoclay and its interactions with biological compounds. The theoretical part deals with the research of the usage of aminoclay as a carrier in medical applications, the method of its formulation, the selection of biologically active compounds and a suitable method of varification. The experimental part deals with the synthesis of aminoclay, samples preparation and study of the adsorption behavior of the selected bioactive compounds under different conditions. The experimental part of the paper also includes varification of the suitability of the selected UV VIS spektrometry to determine the adsorption behavior of the biologically active compounds in colloidal aminoclay solutions.
Fluorescence spectroscopy in the research of aminoclays

Aminoclay belongs to a large group of organic modified materials based on clays. These materials could find many potential practical applications and, in particular, may be used as a matrix with the ability of the organisation and stabilisation of incorporated substances. The utilisation of aminoclay in the field of medical applications seems highly prospective, but there is the need to visualise the clay system and the processes within it. This essay deals with the issue of aminoclay visualisation by means of fluorescent spectroscopy. Through fluorescent spectroscopy, aminoclay was discovered to be autofluorescent. The autofluorescence of aminoclay is not usable in practical applications because of the low fluorescence intensity. This is the reason for the research into other possibilities for modifications of the internal structure of aminoclay by europium ions. These ions in chelated form are able to be fluorescent and modifications of the external structure are possible by electrostatic implementation of fluorescent dyes. Characterisation of the prepared complexes was provided by spectrofluorimetry and fluorescent microscopy for the purpose of evaluation of the practical application of aminoclay.

Theoretical part deals with study of current problems of aminoclay complexes with bioactive substances and the choice of substances for complexing with aminoclay. The experimental part consists of preparation of aminoclay complexes with selected bioactive agents at various concentrations. Verification of binding of bioactive agents to the aminoclay matrix was performed by Elemental Analysis (EA) and Fourier-transform infrared spectroscopy (FT-IR). For finding of the bound amount of bioactive substance were used the Ultra Performance Liquid Chromatography (UPLC®) and analysis of the cytotoxic properties of the formed complexes by used by the MTT assay. The main motivation of this study is to create new complexes with improved characteristics that would replace existing forms of substances used in pharmaceutical and biomedical applications.
